A comparative analysis of the isolates of Clostridioides difficile derived from different sources.
Eretová, Veronika ; Krůtová, Marcela (advisor) ; Ježek, Petr (referee)
Clostridioides difficile infections (CDI) are generally perceived as healtcare associated infections. However, there has been recently reported an increase of CDI incidence in the community. The occurrence of C. difficile has been described also in animals, food and the environment water and soil. The aim of this thesis was to characterize C. difficile isolates derived from different sources using molecular methods. The results were discussed with available data from Czech human C. difficile isolates. A total of 135 C. difficile isolates from the following sources were analyzed: pigs n = 57, calves n = 44, horses n = 18, water n = 15 and hedgehog. Using PCR ribotyping, 22 distinct ribotyping profiles were identified, the most frequently detected ribotypes were: 033, 011, 126, 078. Both toxigenic and non-toxigenic ribotypes were detected, including binary toxin-producing strains. The most frequently detected antimicrobial resistances were to ciprofloxacin, clindamycin, and erythromycin. All ribotypes and sequence types identified in the C. difficile isolates from animals and the environment, has been found also in C. difficile isolates from humans which confirms the role of animals and the environment as a source for C. difficile. However, no epidemic ribotype 001 and 176 that dominate the current...
Cultural houses in the territory of Prague and Central Bohemia in the period 1948 - 1989 and their integrated works of art
Jamnická, Alena ; Alt, Jaroslav (advisor) ; Tourek, Jiří (referee)
The aim of the diploma thesis is to present artistic objects conceived within the architecture of cultural houses since the onset of the communist regime in 1948. The theoretical part of the thesis introduces the development of architectural tendencies of the first and second half of the 20th century, the process of construction of cultural houses and, in context, the mood of the society at that time. The practical part presents a selection of five houses of culture built between 1948 - 1989, which are located in the geographical area of Prague and Central Bohemia. Data collection was carried out using field research methodology, through a critical evaluation of available historical sources and secondary literature. The supporting element of the research is the photographic documentation of the artworks and architecture of the cultural houses, which is accessible within the appendices of the thesis. Keywords Cultural Houses, Art in Architecture, Czech Architecture after 1948, Socialist Realism, Typification, Communism, Prague, Central Bohemia
Analysis of methicillin resistant Staphylococcus aureus strains from Czech Republic and Slovakia
Brajerová, Marie ; Tkadlec, Jan (advisor) ; Karpíšková, Renáta (referee)
Methicillin-resistant Staphylococcus aureus (MRSA) is a major nosocomial pathogen that has spread into the community since the 90s. In general, the community-associated (CA- MRSA) strains are more virulent, but less resistant to antimicrobials than the hospital- acquired strains (HA-MRSA). Some lineages of MRSA such as sequence type 8 have been transmitted more successfully around the world than others and there are situations where a dominant lineage has been replaced by a new one. The factors that are crucial for the selection of dominant lineage are often not clear. As part of this thesis, a longitudinal study of MRSA epidemiology in the Motol University Hospital during the period of 2018-2020 was performed and the occurrence of MRSA clonal complexes was characterized. A multicenter study of the epidemiological situation of MRSA in Slovakia during the period of January - April 2020 was also performed concomitantly. Moreover, several isolates from dominant lineages were further characterized by their phenotype. In the Motol University Hospital study, it was found that the HA-MRSA of the clonal complex 5 dominates and is represented mainly by spa-type t586; a finding that concurs with recent studies from the Czech Republic. In Slovakia, similar to the Czech Republic, MRSA lineages from clonal...

The theoretical section of the thesis characterizes the term institutionalization based on the work of Berger and Luckmann, institutionalization of leisure and there are also some results of previous research into university students' leisure presented. The research section of the thesis consists of own research, for which the quantitative research method in the form of interviews was used (seven respondents).
Typing schemes of two generations, or what's the role of our age in evaluation of strengers in everyday reality
Baranková, Lenka ; Paulíček, Miroslav (advisor) ; Trusinová, Romana (referee)
This work's focus is on research of processes, which we absolutely naturaly and subconsciensely use in everyday reality to evaluate people and situations. It's conserne is describing this proces and furtheron researching the evaluation of unknown people in praxis, mostely from the point of view that evalution can be infuenced by the age of respondent. Main theoretical groung of this work is social constructivism, mostely because we have our statement towards most of all everyday situations and actions just because these exist, therefore are construct of that given society. Furtheron this work focuses on hierarchical organising of these valuating processes, as is typization and categorization and statement based on these. Most of all this work describes differences in evaluating unknown people using photographic interviews of two different age groups of respondents. Data analysis shows, that age and even sex of respondents really does matter in evaluating of unknown people.
